Abstract
Aortaaneurismer utgjør en heterogen gruppe sykdommer med potensielt alvorlige komplikasjoner. For å bedre forstå fundamentale sykdomsmekanismer er gode dyremodeller sentralt. Vi ønsket å etablere en ny metode for induksjon av aortaaneurismer med degenerativ etiologi hos villtype mus ved hjelp av proprotein convertase subtilisin/kexin type 9 (PCSK9) -overekspresjon og Angiotensin II. Vi har derfor gjennomført et pilotforsøk for å undersøke gjennomførbarheten, effekt av behandlingen og mortalitet underveis. Vi har også undersøkt om ultralyd egner seg for å vurdere aortadimensjoner underveis i studien. Rent praktisk har vi har gitt C57BL/6J (WT) mus rekombinant adeno-assosiert viral vektor (pAAV/D377Y-hPCSK9), som koder for et humant, overaktivt PCSK9 gen (D377Y), og angiotensin II (ATII) infusjon i 4 uker. Musene blir gitt høyfett/høy kolesterol diett, som skal etterlikne et vestlig kosthold, ad libitum for å øke blodlipidnivå. Vi har vurdert aortadimensjoner ved hjelp av ultralyd, samt tatt blodprøver for å følge effekten av den virale vektoren på lipidstatus. Metoden har vist seg å være relativt enkel å gjennomføre, med et akseptabelt nivå av mortalitet underveis i studien. Aortadiameteren økte i liten grad, og graden av dilatasjon varierte mellom de ulike avsnittene av aorta. Vi vil derfor vurdere å øke ATII-dosen ved fremtidige studier for å oppnå en større effekt.
Aortic aneurysm, or dilatation of the main artery, is a severe condition with potentially fatal complications. To better understand the fundamental mechanisms of this disease, we have tried to induce aortic aneurysms in mice by combining inducible hypercholesterolemia and hypertension. Furthermore we wanted to assess the feasibility, effect of treatment and level of morbidity/mortality during the treatment. First, the mice were injected with recombinant adeno-associated viral vectors (pAAV/D377Y-hPCSK9) to overexpress a gain of function form of human proprotein convertase subtilisin/kexin type 9 (PCSK9) (D377Y) to increase degradation of low density lipoprotein (LDL) receptor (LDLR) and generate a mouse prone to hypercholesterolemia. After 3 weeks on a Western type diet (high fat/high cholesterol), we implanted miniosmotic pumps with angiotensin II (ATII). The mice received ATII for 4 weeks. To evaluate the effect of the PCSK9/ATII-treatment, aortic dimensions were measured using ultrasound. Hematoxylin/eosin staining and pentachrome staining of aortic biopsies was conducted to evaluate histopathological changes in the aortic wall. The method was relatively easy to use, with an acceptable level of morbidity and mortality during the experimental period. However, the degree of aortic dilation was relatively small and there were differences in aortic dilatation within the study group. For future studies we will consider to increase the ATII-dosage to augment aortic dilatation.
