BACKGROUND:Pituitary adenomas account for about 10 % of intracranial tumours. Theycause morbidity by compression of regional structures or inappropriatesecretion of pituitary hormones. Non-functional pituitary adenomasrepresent about 30 % of pituitary adenomas.The patients experience growth and/or pubertal failure, headaches, visualdisturbances or hypopituitarism from regional compression. Little is known of the oncogenesis of pituitary adenomas
The goal of this study was to detect interesting proteins with are highlyexpressed in patients with non-functional pituitary adenomas, and toexplore them further on.In this project we decided to take a further look at particularly twoproteins, RKIP and NSE.Raf kinase inhibitory protein: RKIP’s ability to influence intracellularsignalling cascades, cell cycle regulation, the suppression of metastasis,neurodegenerative processes, the modulation of emotions, and reproductionhas made it an attractive research candidate for the last decade.Neuron-specific enolase (NSE): Neuron-specific enolase is a substance thathas been detected in patients with certain tumours, namely: neuroblastoma,small cell lung cancer, medullary thyroid cancer, carcinoid tumours,pancreatic endocrine tumours, and melanoma. Studies of NSE as a tumourmarker have concentrated primarily on patients with neuroblastoma andsmall cell lung cancer.We know that RKIP has an important role in suppressing metastasis ofcancer in humans.Our project tried to see if there was any correlation between NSE andRKIP, so that we might use serum NSE to identify patients with a more aggressive non-functional pituitary adenoma.
METHODS:This was done by running 2D-polyacrylamide gel-electroforesis from samples taken from 18patients with non-functional pituitary adenomas. Then we stained the gelswith coomassie blue to detect the spots. All together 20 gel spots in gelsfrom 2D-PAGE were sent to PROBE proteomic unit at university of Bergen foridentification on their protein content.This gel spots were taken from gels that had been stained with coomassieblue colour.Finally we used the Decodon delta 2D program to analyse if there was anycorrelation between RKIP and NSE expression in the tumors. We also run all the same 18 samples with western blot, and then analysed the membranes, without seeing anyclear correlation between these two proteins.
RESULTS:The analysis with the Decodon delta 2D of coomassie blue stained polyacrylamide gels did not show any clear correlation between RKIP and NSE protein content of the tumors.Neither did the analysed membranes from western blot show any clear correlation between those two proteins.
CONCLUSIONS:Despite of getting negative results, we found the RKIP protein sointeresting that we decided to see if low adenoma levels of RKIP correlateto poor octreotide response in acromegaly.The work of this project resulted in a publication in ”The journal ofclinical endocrinology & metabolism”
